Nephalem Valor is a new system introduced in Diablo III. It is intended among other systems to make sure that the end game is fun and exciting, and also that the game is balanced so a 3-minute run of the same area won't be the most efficient way to acquire loot.
Mechanics
Nephalem Valor (NV) is one of the major new systems in Diablo III and it kicks in automatically at level 60. The character being played must be level 60 in order to benefit from the buff. It is not an account-wide system like the Blacksmith or the stash.
Once the player reaches level 60, and kill kills an elite monster pack, they are granted one "stack" of the buff. They can also be granted by opening a Resplendent Chest. The buff stacks up to five times. Each stack lasts thirty minutes, and the buff is removed if the player leaves the game (or is disconnected), changes skills, or changes acts. Killing an elite pack will refresh the time on the buff, and add a number to the stack if it isn't at maximum.
The buff itself increases Gold Find and Magic Find by up to 75% (at fifteen percent per stack). However, that is a minor bonus compared to what the player gets upon killing other elite enemies: more loot.

When the player is at a full stack, (or in other words five stacks of NV), they will receive more Magic and Rare items from other elites. There are exceptions to this, however.
NV stacks are only awarded by rare or champion packs of elite monsters, whose names appear in blue or yellow. When killed, they will drop loot and also award one thirty-minute stack of NV, or else they will refresh the time on the stack. The player will then receive more loot from other champions and rares that they kill, and some Superuniques (who are always quest-related bosses). Superunique monsters, whose name appears in purple, do not drop more items per the Nephalem Valor buff, although their drops are affected by the gold and magic find that comes with the buff.
At a full stack, a boss monster is mostly guaranteed to drop two rare items and on average six magical items, in addition to the customary gold drops (which is increased in quantity via NV's gold find boost).
