The Key of Destruction is a special item dropped by Odeg the Keywarden, who can be found somewhere in the Fields of Misery in Act One.
On Torment difficulty this monster may drop the Key of Bones; one of the four keys required to create the Portal Device, which is required to complete the Infernal Machine event.
